The Barcelona defender tore the abductor muscle in his left leg in Saturday's 1-1 draw with Deportivo La Coruna and had to be replaced midway though the second half.
He is now expected to be sidelined for around two weeks, making him doubtful for the competition in South Africa.
Spain coach Vicente del Bosque is set to name his squad on Monday and has a number of other fitness concerns.
