Wolves made the brighter start to the match, with Jemal Johnson's pace, in particular, worrying the hosts, but as the half wore on, The Whites increased the pressure on Mick McCarthy's team.
Ian Moore twice drew good saves from Matt Murray, and defender Paul Butler volleyed wide.
Ten minutes before the interval, Leeds missed a gilt-edged opportunity to take the lead.
A cut back from Jonathon Douglas, making his first start since rejoining the Elland Road club, found Sebastien Carole plum centre of goal, who seemed odds-on to score, only for Murray to block from point-blank range.
Leeds would have been disappointed not to have registered at least one goal for their first-half endeavours, but Kevin Blackwell's side almost found themselves trailing within seconds of the restart.
Half-time substitute Jay Bothroyd was the creator, he teed up former Blackburn Rovers team-mate Johnson, but the livewire could not beat Tony Warner at his near post.
Play swung this way and that in the second period. Eddie Lewis put a free kick wide, while Johnson, who later went off through injury, found the side-netting.
Substitute Jermaine Beckford's cross caused confusion between Murray and Jody Craddock late on, and Gary Breen saved Murray's blushes after the goalkeeper flapped at a cross, as Wolves appeared happy to settle for a point.
But, just as the match appeared destined for a goalless draw, Bothroyd pulled a finish right out of the top drawer - turning Douglas 25 yards from goal, he unleashed a shot which found the top right-hand corner to hand the visitors all three points.
