Top seed and world No 1 Williams will continue her quest for a sixth Australian Open title - but her first since 2010 - when she clashes with Slovakia's Daniela Hantuchova, ranked 31.
Williams tied Margaret Court on 60 career wins in Melbourne and will be looking to take the record outright against Hantuchova, who overcame Britain's Heather Watson in the first round.
Djokovic, who is bidding for a fourth successive title in Melbourne, will be last on in the night match against Denis Istomin of Uzhbekistan as the third round gets underway.
The Serbian holds a 3-0 head-to-head record over Istomin and claimed a 6-1 6-1 6-2 victory over his opponent at the same stage of the tournament in 2010.
Djokovic has not dropped a set in his first two matches in the Melbourne heat and will be looking for a repeat performance on Friday with the boiling conditions set to continue.
Also on the main showcourt, men's third seed David Ferrer will take on Jeremy Chardy of France, ranked 29th.
The pair last played at Wimbledon four years ago when Ferrer came through a fifth set decider 7-5, and with both players dropping a set in second round wins it could be a closer match than the rankings suggest.
Popular Aussie Sam Stosur is also on Rod Laver Arena as the 17th seed takes on 14th ranked Ana Ivanovic, while Casey Dellacqua is another Australian handed the privilege of playing on the big stage when she plays Zie Zheng of China.
Over on Hisense Arena women's ninth seed Angelique Kerber kicks off the action against American Alison Riske, while fourth seed Li Na takes part in the following match against Lucie Safarova of Czech Republic, the 26th seed.
Seventh seed Tomas Berdych ought to have few problems against Bosnian Damir Dzumhur, a qualifier ranked 188 who bested Ivan Dodig of Croatia in four sets in the second round.
Match of the day on Hisense could be 17th seed Tommy Robredo of Spain versus ninth seeded Frenchman Richard Gasquet, whose four previous matches have been shared.
There is also some good singles action on Margaret Court Arena where 19th seed Kevin Anderson takes on Edouard Roger-Vasselin and 15th seed Fabio Fognini (Ita) meets big-serving American Sam Querrey.
