Ulster scrum-half Ruan Pienaar comes into the starting XV in place of Jano Vermaak, who sustained an injury during last weekend's 44-10 rout over Italy.
The only other switch in the starting line-up sees Marcell Coetzee deputise for the injured Francois Louw at blindside flanker.
Prop Bismarck du Plessis makes a welcome return to Test rugby on the Springboks bench after 10 months out with knee injury, while head coach Heyneke Meyer could hand debuts to Siya Kolisi and Piet van Zyl who are both named in the match-day 23 for the first time.
Meanwhile Scotland coach Scott Johnson has named an uncapped trio in his side as Scotland look to bounce back from their opening round defeat to Samoa.
Full-back Peter Murchie, winger Tommy Seymour and lock Tim Swinson will all make their international bows at the Mbombela Stadium, as Johnson makes seven changes to the side which lost 27-17 last Saturday.
Ruaridh Jackson makes his first appearance in Scotland colours since the Six Nations as he replaces the concussed Tom Heathcote at fly-half.
Injuries have also forced Johnson to makes changes in the pack with hooker Pat MacArthur replaced by Scott Lawson while flanker Ryan Wilson deputises for captain Kelly Brown.

What they said...
South Africa head coach Heyneke Meyer
"It's great to have Bismarck back in the mix, but I'm even more excited about the inclusion of Piet and Siya on the bench.
"This time last year, Piet and Siya, along with Arno Botha, were part of the group of six young guns who trained with us and now all three are in the Test match squad."
Scotland coach Scott Johnson
"We've faced South Africa a fair bit in the last decade and right now they're the second best team in the world, playing on their own paddock, and with a win against Italy already under their belts.
"We have to be competitive and, once again, I'll reiterate that we have to get our accuracy and aggression 100% at breakdown to give ourselves the opportunity."
