Yohan Blake, silver medallist in both the 100m and 200m at London 2012, led home the quartet in a time of one minute 18.63 seconds to beat the old record by 0.05secs, which was set by the USA in 1994.
Nicekl Ashmeade, Warren Weir and Jermaine Brown also played their part for Jamaica, with Brown and Blake completing the final lap of the track in a time of 39.63secs.
St Kitts and Nevis finished second in a national record time of 1min 20.51secs, while France took third and established a new European record of 1:20.66.
Outside the zone
The United States had originally claimed bronze, but their second exchange, from Curtis Mitchell to Ameer Webb, was ruled to have been completed outside the zone.
Meanwhile, Great Britain's men topped their heat in the 4x400m to qualify for Sunday's final in a time of 3:00.74, which the United States and Jamaica could not better in their respective heats.
Christine Ohuruogu is set to come in for the women's 4x400m final on Sunday after Eilidh Child, Shana Cox, Emily Diamond and Margaret Adeoye qualified for the showpiece, despite finishing third in their heat behind the United States and Jamaica in 3:27.30.
