The former England international joined the Welsh region earlier in February and has been named at inside centre for the visit of Alan Solomons' side.
Barkley is the only change in the backs from the last clash the Scarlets played in - their contest against Ulster last Friday having been postponed.
The other alterations come in the pack, with Emyr Phillips and Samson Lee coming into the front row and Johan Snyman being named at lock.
"Olly brings a lot of experience to our squad both from his distinguished test and club career in England," Scarlets head coach Simon Easterby said.
"He adds much needed cover for us as we have so many players unavailable during the international call ups as well as some long term injuries we have in midfield."
Edinburgh boss Solomons has opted not to make any changes to the side which was beaten 11-7 by Connacht last time out.
The only changes to the 23-man squad see front-row forwards George Turner and Ewan McQuillin come on to the bench for Ross Ford and Geoff Cross, who are both on international duty with Scotland.
