The Australian full-back missed his side's last three games including the Easter programme, which saw Hull beaten by their city rivals Hull KR before responding with a win at Wakefield.
Jamie Shaul scored two tries on his Super League debut against Wakefield and retains his place in Hull's 19-man squad - but is likely to drop to the bench to accommodate McDonnell in the starting line-up.
Forwards Paul Johnson and Ben Galea both sit out after playing three games in 10 days, meaning second rower Dean Hadley is in contention for his debut.
Trio injured
Catalan Dragons recall wing Frederic Vaccari and scrum-half William Barthau, who have both made just one appearance so far this campaign.
Versatile Kevin Larroyer also makes the 19-man squad but Damien Blanch, Daryl Millard and Thomas Bosc all miss out through injury.
The Dragons sit fifth in the Super League table, four places and five points above Hull, but lost their unbeaten home record to 12-man Leeds Rhinos on Easter Monday.
