Mitchell, who was eventually run out to bring an end his 106-ball stay, was well supported by Thilan Samaraweera (41) in a third-wicket stand of 111 and Gareth Andrew added a quick 19 later on to ensure a formidable total.
Seamer Chris Liddle was Sussex's most effective bowler with 3-37 from his eight overs.
Sussex were dismissed for 154 in reply, Worcestershire seamer Jack Shantry finishing with figures of 3-29.
Michael Yardy's 45 was the top score of an innings lasting 31.5 overs and featuring two run outs.
