Knox carded an impressive six-under 64 over the Monterey Peninsula course to join Hunter Mahan at the top of the leaderboard.
It could have been so much better for Knox, though, following eight birdies, however he also dropped two shots.
Mahan birdied five of his final seven holes to finish with a six-under 66 on the Pebble Beach course.
Knox and Mahan hold a one-shot lead over a six-strong chasing pack in California, including England's Greg Owen.
The 40-year-old had a blemish-free round over the Monterey Peninsula course to remain in contention alongside Matt Every, Ted Potter Jr, Scott Langley, Ryuji Imada and Seung-Yul Noh.
Another Englishman, Lee Westwood, is a further stroke behind after signing for a round of 68 at the end of his round on the Pebble Beach course.
The world number eight hit 10 of 14 fairways and 16 of 18 greens in regulation on his seasonal PGA Tour debut.
"My game is in good shape and there's a lot of weeks that I have not been to in the last few years that I'm looking forward to going back to again, weeks like this, Arnold Palmer's tournament, Bay Hill, the Memorial are all on the schedule," he told the PGA Tour's official website.
"So it's going to be an exciting year and hopefully a big year."
