The Gelredome side went into the game holding a five-point lead over Sparta Rotterdam with two games remaining in the play-offs.
Sparta could have taken the group down to the wire with a win, but they were held at home by Helmond Sport and this allowed Vitesse to take top spot.
Matthew Amoah was the man who took the glory for Vitesse, as he netted their goal on 29 minutes following good approach play from Aleksandar Rankovic.
Venlo could have been forgiven for not putting it all in, but they battled to the end and were rewarded with a leveller from Ken Leemans in the last ten minutes.
Rick van den Bergh scored twice for Sparta, but it was in vain as Helmond Sport held them to a 2-2 draw. Rob Oomens and Danny van Bakel were the men on target for Helmond.
Group B is exceptionally tight, with De Graafschap level on points with Heracles at the top with just a game left to play.
Heracles started the day two points clear, but they were forced to come from two down to draw 2-2 with Excelsior.
Rene van Dieren and Mounir el Hamdaoui had Excelsior cruising with 16 minutes remaining, but Heracles dug deep and goals from Richard Roelofsen and Yuri Rose salvaged a point.
Heracles are level with De Graafschap after the latter thumped sorry Volendam 4-0.
A brace from Jhon van Beukering and goals from Milan Berck Beelenkamp and Hilmi Mihci were enough to see De Graafschap home and set up a thrilling final day.
De Graafschap travel to Excelsior on Thursday, while Heracles visit Volendam.
