Danny Hoosen gave Ajax an ideal start when he struck after just two minutes, although there were no further goals in the first half.
Viktor Fischer doubled the lead 11 minutes after the break with Davy Klaassen rounding off the scoring after 79 minutes.
It gave the capital club a one-point lead over Vitesse at the head of the table, with the former leaders playing Go Ahead Eagles on Sunday.
A last-minute goal by Vasil Skourtai saw Roda JC secure a 2-2 draw at nine-man AZ Alkmaar, where they denied the high-flying hosts the opportunity to go to the top of the table - even if only temporarily.
It was a fractious match, with the hosts reduced to nine men after just 19 minutes after Nick Viergever and Nemanja Gudelj were both sent off.
AZ still managed to take the lead through Mattias Johansson (25) only for Wiljan Pluim to level four minutes later.
However, Aron Johansson restored AZ's lead six minutes before half-time and they seemed to be heading to the top of the table only for Skourtai to have the final say.
Roda, though, were reduced to 10 men in injury time when Bert Biemans was sent off.
An injury-time goal by Florian Jozefzoon earned PSV Eindhoven a 1-1 draw with 10-man Heerenveen at the Philips Stadion.
It was unfortunate on the visitors, who were forced to play with 10 men for all but 15 minutes of the match after Joey Van Der Berg was given a straight red card.
They managed to hold off their hosts and even took the lead through Yanic Wildschut 10 minutes from time.
It appeared that they were heading for three points before Jozefzoon's late intervention secured a point.
NEC are still rooted to the bottom following a 2-1 defeat at Cambuur Leeuwarden.
Mohammed El Makrini gave the hosts the lead (33) and although Kevin Conboy equalised four minute later, a Tobias Haitz own goal with a little under 20 minutes to go ensured Cambuur secured all three points.
