Rafael van der Vaart was the star man for the Amsterdam-based outfit grabbing a hat-trick as the hosts completely outclassed their fourth-placed opponents.
Wesley Sneijder got the ball rolling for Ajax in the 17th minute, and van der Vaart got his first just two minutes later.
The Dutch international struck again on the stroke of half-time and completed his treble with eight minutes remaining.
Petri Pasanen and Hatem Trabelsi also struck in the second-half to complete the rout, while Jochen Janssen netted a late consolation for the visitors.
Leaders PSV were forced to come from two goals down at the Philips Stadion to claim a share of the spoils against AZ Alkmaar.
Strikes from Tim de Cler and Ali Elkhattabi handed AZ a two goal advantage at the break, however the game turned on the hour mark when the visitors had Michael Larney sent-off.
PSV stormed forward and a brace from Arnold Bruggink (69, 80) ensured they took something from the game.
Heerenveen inflicted a 5-1 home thrashing on Willem II with John de Jong and Stefan Selakovic both grabbing doubles.
Mike Nurmela also notched for the visitors while Tarik Sektioui scored a late reply for the hosts.
FC Zwolle gave their prospects of avoiding relegation a massive boost after dispatching Uefa Cup chasing Roda JC 3-1 at the Opsterenk Stadion.
Marco Roelofsen and Marino Promes put Zwolle two up inside the first quarter of an hour and despite Roda finding a way back into the game courtesy of an Ivan Vicelich strike, a further effort from Morten Karlsen in injury-time wrapped up the points for the hosts.
Vitesse Arnhem also gave themselves renewed hope of escaping the drop with a 3-1 home win over NAC Breda.
A double from Matthew Amoah (19, 57) and a Bob Peeters effort was enough to see off NAC for whom Orlando Engelaar netted.
