The Paraguay international has agreed terms with the Serie A club after previously being linked with Benfica.
NEC were happy to thrash out a deal for the midfielder, who had 12 months remaining on his contract.
The Dutch side were keen to raise funds for their own forays into the transfer market and general director Jacco Swart feels it is a good deal.
"He has played here for three-and-a-half years," Swart told the Dutch media. "This is a nice step for him, a big European competition."
Barreto joined NEC during the 2003/04 season from Cerro Porteno, but will now try his hand in Italy next term.
Meanwhile, NEC have also confirmed that Polish striker Andrzej Niedzielan has officially completed his move to Wisla Krakow on a three-year contract.
NEC are also looking to add to the squad, with Feyenoord youngster Tim Vincken a target of coach Mario Been.
Vincken made 23 appearances in the Eredivisie last season, but the winger could be squeezed out by new signings at De Kuip.
Been told De Gelderlander: "It's no secret that I am a fan of Tim Vincken. Who knows?
"There will be possibilities within a couple of weeks."
