The Argentine is keen to be reunited with his former Roma team-mate following his big money move to Madrid in the summer.
There was much speculation that Totti would follow Samuel to the Galacticos this summer, but the Italian playmaker chose to stay with the Giallorossi.
Despite this Samuel is hopeful that Totti will eventually join him in Spain.
He said: "It is difficult for Totti to leave. In Rome he is like the Pope.
"But if one day he decides to leave I am sure there will be a place for him here."
Samuel was speaking on the eve of his side's UEFA Champions League clash with Roma.
It will be Samuel's first game against Roma since his switch and he is looking forward to challenging his old team-mates.
"The match will be difficult for both teams.
"I have mixed feelings about playing my old team as I had some very happy moments there," he told Corriere dello Sport.
Both sides have lost their coaches following disappointing starts to the season and are under enormous pressure to get a result.
