The Argentine centre half has been a long-term target of the Spanish giants and he will arrive at El Santiago Bernabeu on July 1.
Samuel, who was also wanted by big-spending Premiership side Chelsea, has concurred on a five-year contract with Real.
He had stated that Madrid was his preferred destination ahead of London and his wish has been granted after Roma reached an accord with Real.
Roma's financial situation had prompted the sale although the size of the transfer fee has yet to be revealed by either club.
Real's sporting director Jorge Valdano is delighted with the acquisition of his compatriot and feels Samuel will be the perfect addition to the team's back-line.
"We have reached with Roma and the player an agreement in principle and, finally, we can announce the signing of a central defender, something we have considered a high priority for next season," Valdano told the club's official website.
"Now we have something pretty to think about.
"He brings a combination of quality and maturity that is going to help our young players to feel more comfortable.
"I understand that he covers an important necessity in the team and, in addition, it is a great thing because he is one of the best defenders in the world."
