Substitute Chris Iwelumo grabbed an equalizer 13 minutes from time to cancel out Steve McPhee's 63rd minute opener.
Vale took charge early on although they had to wait until after the hour mark to open the scoring at Vale Park when McPhee headed in from Stephen Brooker's cross.
The visitors then made it all square when Iwelumo latched onto Lewis Neal's pass and produced a calm finish to hand Stoke a share of the spoils and moves them up into fourth spot in the Second Division.
Cardiff striker Robert Earnshaw grabbed his fourth goal of the season at the County Ground as he gave his side the lead in the 36th minute.
Jason Bowen then doubled the lead for the Welsh side a minute after half time when he made the most of a mistake from Babby Howe to make it 2-0.
Roy Evans's side then had David Duke sent off on the hour to effectively kill the game off for The Robins.
Cardiff skipper Graham Kavanagh then sealed a good win for Alan Cork's men with a powerful header in the 80th minute to give them their first win in six outings.
