The 17-year-old, who made his debut for Derby at just 15 to become the club's youngest ever player - has agreed a two-year contract, the maximum for a player of his age.
"It's great to be signing for a club like this with great potential," Holmes told the club's official website.
"I joined Derby as a kid and it was my dream then to play for the first team - it's great for me that it has come true.
"You have to stay with your roots and my mates from the old days are still with me - they are great lads.
"Living close by, I have always had Derby at heart. I made the records at a young age but this year has been an incredible learning curve and I never thought at the start of the season I would start so many games.
"Tom Huddlestone, Nathan Doyle and myself have all broken through to the first team and there are at least a couple more from our intake at the academy who I have faith in."
Director of football Murdo MacKay confirmed Mawene was set to leave the club.
"Youl is obviously examining his options and to my mind, that means we are no longer his first choice," said MacKay.
"That is unfortunate because right now we want players who are committed to this football club.
"Youl's people are well aware of the terms that have been offered but it is becoming increasingly difficult to talk to them, and we have had no success whatsoever in speaking to Youl himself.
"That is particularly disappointing because this club has been good to Youl, particularly during his spells out of the game through injury.
"I have spoken to George Burley about the situation and we both agree it would be best all round if the player went elsewhere.
"The likes of Richard Jackson, Adam Bolder, Junior and Lee Holmes all had options, but they decided to stay.
"It sends a negative message out to the squad when it is perceived that one particular individual is playing for time.
"If he isn't going to sign then, with respect, we need to use that time to find a replacement.
"We believe with the money on the table for Youl, we can bring in a top class defender."
