Reyna was stretchered off in Monday's 1-1 draw with Bolton after he fell awkwardly and now scans have confirmed the Black Cats' worst fears.
US national team orthopaedic consultant Dr. Bert Mendelbaum, flew to England to deliver the news to Reyna and discuss what will happen next.
Reyna's knee is still swollen and so surgery cannot be carried out for at least another two weeks, although he has been booked in for the operation in Los Angeles already.
"It's always a big blow to lose good players but unfortunately that's life," said Sunderland boss Howard Wilkinson.
"Claudio's problem is someone else's opportunity. His credentials are there for all to see but for the immediate and mid-term future he will not be available, and that's a blow."
Sunderland are already without the likes of Thomas Sorenson, Thomas Myhre and Jason McAteer and Reyna's injury woe is just the latest setback for the Wearsiders.
Yet more bad news could follow over the next few days, after Argentine star Julio Arca was carried off in the reserve team encounter with Manchester United on Thursday evening.
Arca has damaged his ankle and could face a prolonged spell on the sidelines, although the club will have to wait for X-rays this weekend before learning the extent of the injury.
