Orient midfielder Chambers has started every League One game this season and has now committed his future to the club until 2010.
His decision to stay with the O's has delighted manager Martin Ling, who signed the 27-year-old on a free transfer 18 months ago.
"Adam is a massive part of what we are trying to build at this club, so it's excellent news that he's signed this new contract," Ling told the Os' official website.
"He has been with us 18 months now and is certainly one of the first names on the teamsheet every week.
"He is a quality player for us and one that we can build around."
Meanwhile, Spurs striker Barcham has extended his loan with Orient until the end of the season, while Sam Oji is close to rejoining the club.
Barcham has yet to score in five appearances for Ling's men since initially signing in late November.
