John Terry and Frank Lampard scored but Chelsea were beaten 3-2 by the Major League Soccer All Stars on Wednesday evening in Pennsylvania.
Terry and Lampard's goals had put the UEFA Champions League winners ahead early in the second half after Chris Wondolowski's opener.
But an MLS All Stars team that included David Beckham and Thierry Henry equalised through Chris Pontius before sealing a dramatic victory in the final moments.
With the game heading for a penalty shootout, former Fulham forward Eddie Johnson netted with a deflected effort in injury time to inflict Chelsea's first defeat of their North American tour.
Marko Marin played the first hour for the Blues, while fellow new boy Eden Hazard came off the bench for the final 30 minutes.
Roberto Di Matteo also included Yossi Benayoun in his starting line-up as well as Romelu Lukaku, who operated as a lone target-man.
The defeat comes after Chelsea beat Seattle Sounders in their tour opener before drawing with Paris St Germain. They now face AC Milan in Miami on Saturday.
