It was the 10th successive wins for the Broncos, who now move in to a tie for top spot in the AFC standings alongside the Houston Texans.
Peyton Manning was at his inspirational best as he completed 30-of-43 passes for 339 yards and three touchdowns.
Manning connected with Demaryius Thomas and Eric Decker (twice) to throw three touchdown passes for the 72nd time of his career.
Manning and Thomas linked up from 22 yards for the opening score - with the Browns responding with a field goal from Phil Dawson.
It was 14-3 at the break, though, as Manning's 10-yard pass to Thomas put the Broncos in control.
Dawson's second field goal kept the Browns in touch, however Decker and Manning combined again - this time from eight yards.
And in the final quarter Matt Prater slotted his first field goal and Jacob Hester rushed over from a yard to put the result beyond doubt.
Greg Little went over for Cleveland's only touchdown but Denver had the final word, courtesy of Prater's second field goal.
